Well, I'll try to analyse my answer as much as I can.
Simba: Simba was mentally scarred by the events of the stampede, his father's death and Scar's accusation of him (Simba) being a murderer. He seemed too young at the time to think about the events more clearly/suspect Scar, or simply tried not to think about them. He lived with his regret even in his life in the jungle that followed him as an adult. As an adult he had to face his greatest fear, which was facing his mother/pride and become King. As a father he's filled with angst and insecurity as Zira and the Outsiders are around and threatening to take over the Pride Lands and he still has nightmares about his father's death so it's obvious he hasn't had a peace of mind even since he defeated Scar and his insecurity results in the overprotection of Kiara and distrust of Kovu. At the end of SP he had to face Zira and defend the Pride Lands. After defeating Zira and uniting the prides, he can finally rest for once in his life.
TL;DR He lived with guilt and insecurities all his life.

Nala: Nala had to face the supposed death of her friend, Simba and survive through Scar's reign without a fatherly figure (or a friend to grow up with, because no other cubs were shown in the movie). Scar's ruling was so terrible that as an adult, Nala had to leave the Pride Lands and find help. At the end of both movies, she's secure and laid back.
TL;DR She was mostly alone while growing up and had to endure Scar's ruling.

Kiara: Because of Simba's insecurity, Kiara had to grow up alone and always with her babysitters when all she wanted was to be free so she could never be truly happy during her childhood. As a young adult, she had to convince her father that Kovu wasn't evil and had to face her father. In the end of the movie she marries Kovu and unites the prides, meaning she can finally have a somewhat more peaceful life.
TL;DR She had to endure her father's over-protectiveness.
Kovu: Kovu was raised in hatred with only one goal in his life: Kill Simba and take over the Pride Lands. He suffered through intense training by Zira while living in the harsh conditions of the Outlands and barely had any affection by his mother at all. As an adult, he was filled with hatred until he met Kiara and after he changed his mind about the Pridelanders and had to face his pride/mother and Simba's judgement. In the end of the movie he marries Kiara and unites the prides and is finally at peace.
TL;DR Extremely harsh childhood and hesitating mental state as an adult.
Nuka: Nuka was ignored and treated like trash all his life. Sadly we know very little about him. As a teen, he's seen as a filthy termite-infested lion that wants his mother's attention. As an adult, he's really bitter towards Kovu and completely desperate to impress his mother, resulting to his death after attempting to kill Simba.
TL;DR Was treated like nothing his entire life.
Sarabi: Sarabi's childhood is unknown, but as an adult we do know she suffered through Scar's reign while possibly trying to cope with both her husband's and son's (supposed) death, which was truly tragic.
TL;DR Had to cope with the loss of her family members and Scar's reign.
My conclusion: After analysing all this, I believe that the one with the worst life must be... Nuka. Being treated like trash and ignored like that is just mental torture and possibly the worst kind of torture. None of the other characters had to face that. (Well, Kovu/Simba are close to this one, but still distant compared to Nuka's life.)
