She knows Simba is hiding something ("He's holding back, he's hiding") so she starts to beleive Scar and pay more attention to his words. The other lionesses know nothing about Simba so they're not beleiving Scar at all.
Scar is looking at Nala because he knows she was the one who brought Simba back. Nala was the one who defended Simba ("Simba is the rightful king") to begin with. He knows she's close to Simba and is using his feelings for her to screw with his mind.
