I really didn't think about that, I always saw it as just ordinary grass with dust, nothing more

He probably grabbed the "stuff" because it came from the sky and he reads omens. And then he smelled Simba

I never understood why he grabbed the dust and stuff in the first place, but I think he knew by the scent or it had small bits of Simba's fur in it.

Rafiki might grab random grasses that float bye all the time as a form of news gathering. He didn't pay any particular attention to Simba's patch of grass until he realized that it had Simba's scent on it, so it really looked like something he does on a regular basis.

It's like reading the newspaper, and suddenly reading some great news. You don't know that the paper is special, and treat it as another news source. Only when you finally read it do you realize that it held an important message.
