For me I wanna say the scene where Mufasa comes back to tell Simba he needs to take his place as "The One True King" and to "Remember who you are"
What about you? :D

So many...

The Lion King:
I agree with juhouh that The Circle of Life was a power opening.
Under the Stars\ The Great Kings of the Past.
The Stampede
I agree with you about Mufasa's ghost telling him to go back.
The fight with Scar.
His roar in the rain.
The ending with Kiara's presentation.

Simba's Pride:
We are One
Love will Find a Way
Nuka's death
The We are One speech

The Lion King 1 1\2:
Timon deciding to leave
Timon deciding to go back to the Pride Lands to help.
Simba saying he couldn't have done it without them.
Timon risking his life to dig a hole for the hyenas to fall into.

It depends on how you define "powerful"; personally, I define it in terms of a movie as a scene that is the most uplifting and inspiring. So, I select Mufasa's ghostly visitation with Simba as the most powerful scene in the movie. Over the years, I have read comments that people admitted to crying throughout the scene, but it uplifts you to become greater than one's self.

Like others have said already, Mufasa's Ghost is my choice, too. I see it as the turning point of the film - it is the part where Simba finally decides to take his destiny seriously and stop running from his past. In a way it is the completion of Simba's character arc, since as we all know he goes back to Pride Rock after that, and defeats Scar with a surge of determination given to him by this encounter (and the fact that he now knows the thing he has spent his whole life feeling guilty about wasn't even his fault, after all). Though, really, if one of my relatives appeared to me in a giant-ass storm cloud I'd probably do whatever they told me to do, too
